Advanced On-bead DEL Technology for Enhanced Drug Discovery Screening

DNA-encoded library (DEL) technology has been recognized as a major screening method with unique advantages for offering vast chemical diversity and multiplexed affinity-based screening. Recently, solid-phase On-bead DEL, also known as one bead one compound (OBOC), has been developed to further expand DEL screening from affinity-based screening into biochemical activity screening.

At the 2025 SLAS conference in San Diego, WuXi AppTec presented a poster showcasing a multiple application case study of OBOC, including biochemical activity screening and traditional affinity screening. For biochemical screening, with high-encapsulation rate microfluidics chip design, the authors demonstrate successful screening of a 200K-member OBOC library. In affinity screening, the authors showcase how molecular glues can be screened using a special focused library coupled with affinity-based screening methods.
