Mechanistic Profiling of ADC-resistant Cancer Cell Lines

Antibody-drug conjugates (ADCs) have emerged as an important modality in targeted cancer therapy development. However, acquired resistance continues to limit their long-term clinical efficacy, underscoring the need for robust preclinical models to better understand resistance mechanisms.
At AACR 2026, WuXi Biology presented a poster entitled “Systematic Mechanistic Profiling of ADC-Resistant Cancer Cell Lines.” Through chronic, escalating ADC exposure, our scientists successfully established several ADC-resistant cancer cell lines, with most models exhibiting more than 10-fold increases in IC50 compared with their parental counterparts.
This work highlights the diverse mechanisms underlying ADC resistance across different cancer types and cell lines, supporting the discovery of new strategies to overcome ADC resistance in cancer therapy.
